摘要
目的 探讨吲哚氨2,3-双加氧酶(IDO)及犬尿氨酸氨基转移酶(KAT)活性在脑梗死患者中的变化情况及病理作用.方法 正常对照组35例和脑梗死组81例,采用免疫透射比浊法测定血清超敏C反应蛋白(hsCRP)、载脂蛋白A1(APOA1)、载脂蛋白B(APOB)浓度,采用酶法分析甘油三酯(TG)、总胆固醇(CHO)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL)、游离脂肪酸(NEFA)浓度,采用HPLC-荧光衍生法测定血液色氨酸(TRP)、犬尿氨酸(KYN)及犬尿喹啉酸(KYNA)浓度,计算IDO及KAT比活性,并进行对照分析.结果 脑梗死组TRP、KYNA、HDL、KAT比活性明显低于对照组(P均<0.05).脑梗死组hsCRP、IDO比活性明显高于对照组(P均<0.01).脑梗死患者IDO比活性与hsCRP呈正相关(r=0.425,P=0.027).结论 脑梗死患者体内存在炎症反应,炎症应答上调IDO的活性表达,进而影响KYN代谢途径,可能与脑梗死患者的病理生理密切相关.
Objective To investigate the changes of indoleamine 2, 3 -dioxygenase (IDO) and kynurenine aminotransferase (KAT) in the patients with cerebral infarction and it's pathophysiological role in disease progress. Methods A total of 35 cases of healthy adults and 81 cases of cerebral infarction patients were recruited in this study. The level of high - sensitivity C - reactive protein( hsCRP), APOA1 and APOB in serum were analyzed by immune transmission turbidity method; TG, CHO, HDL, LDL, NEFA levels were determined by enzymatic method. We applied HPLC - fluorescence derivatization method to the determination of tryptophan ( TRP ), kynurenine ( KYN), and Kynurenic acid(KYNA) concentrations in serum. The IDO and KAT ratio activity were denoted by KYN/TRP ratio and KYNA/KYN ratio, respectively. The correlation between hsCRP and IDO, KAT were analyzed. Results TRP, KYNA, HDL and KAT ratio activity was significantly lower in the cerebral infarction group than in the control group ( all P 〈 0. 05 ). HsCRP and IDO ratio activity was significantly higher in the cerebral infarction patients than in the control group ( both P 〈 0.01 ). IDO activity in the patients with cerebral infarction showed a positive correlation with hsCRP (r =0. 425 ,P = 0. 027). Conclusion There is inflammatory response in the patients with cerebral infarction in vivo, which increases the activity of IDO and affects the KYN pathway, may be related closely to the pathophysiotogy of patients with cerebral infarction.
