摘要 目的:研究微波辅助萃取马铃薯叶中茄尼醇的最佳工艺条件。方法:以95%乙醇为溶媒,高效液相色谱法测定提取液中茄尼醇的含量,采用L16(44)正交设计法考察料液比、萃取时间、微波辐照功率和萃取温度四因素对茄尼醇提取率和浸膏得率的影响。结果:4个因素对马铃薯叶中茄尼醇提取工艺的影响顺序依次为萃取温度、料液比、微波辐照功率、萃取时间,其中萃取温度对提取工艺有显著性影响(P<0.05)。结论:用95%的乙醇作溶媒微波辅助萃取马铃薯叶中茄尼醇的最佳工艺条件为料液比1∶8(g.mL-1),提取时间20min,提取温度65℃,微波功率450W。在此条件下茄尼醇的提取率为94.19%。 OBJECTIVE To optimize the microwave assisted extraction (MAE) process of solanesol from discarded potato leaves by orthogonal method. METHODS 95% ethanol was used as extraction solvent, the contents of solanesol was deter mined by the reversed phase high performance liquid chromatography (HPLC), and L16 (4^4) orthogonal testing method was used to its essential step like ratio of material to solvent, extraction time,microwave radiation power and extraction temperature so as to analyze the factors which influenced the extraction yield of solanesol by MAE. RESULTS The order of factors which influenced the extraction yield of solanesol by MAE was extraction temperature, ratio of material to solvent, microwave extraction power,radiation time,and extraction temperature had a significant effect on MAE(P〈0. 05). CONCLUSION The results of orthogonal test indicated that the optimum microwave assisted extraction conditions of solanesol from discarded potato leaves were chosen as volume fraction 95 % alcohol as solvent, ratio of material to solvent 1:8 (g · mL^- 1 ) ,microwave radiation power 450 W, temperature 65℃, extraction time 20 minutues. The yield of solanesol was 94. 19% on the optimum microwave assisted extraction condition.
