摘要
目的研究硫酸依替米星单次给药在大鼠肾脏中的药代动力学。方法大鼠单次肌肉注射2mg/kg硫酸依替米星,分别于给药后0.5、2、4、8、12h和1、2、4、7、10d断头取血,同时剖取肾脏,用高效液相色谱法测定血浆和肾匀浆中硫酸依替米星的浓度。结果给药后2h血浆中硫酸依替米星的浓度己降至(0.33±0.1)μg/ml。肾脏中硫酸依替米星的浓度为(31.98±5.69)μg/g,在肾脏中的总量是(50.69±15.92)μg,接近峰值水平,约占给药量的14%;给药2d后肾脏中的依替米星开始下降,4d后下降约50%,7d后下降约85%,10d后除有1只大鼠的肾脏中尚能够检测到硫酸依替米星外,其余实验动物肾脏中均未检测到硫酸依替米星。结论大鼠单次肌肉注射硫酸依替米星能够在肾脏中明显蓄积。
Objective To investigate the properties of the accumulation of etimicin sulfate in the kidney of rats by a single-dosage administration.Methods After the delivery of etimicin sulfate by a single-dosage intramuscular injection at the dosage of 2 mg/kg,blood samples were taken from the rats by decollation at 0.5,2,4,8,12 h and 1,2,4,7,10 d,and then the rats' kidneys were taken out by anatomy.Five rats were examined at each point of time.The concentration of etimicin sulfate in plasma and kidney samples was determined using high performance liquid chromatography.Results Two hours after administration,the concentration of etimicin sulfate in the plasma decreased to(0.33±0.1)μg/ml while the concentration and the quantity of etimicin sulfate in the kidney of the rats were(31.98±5.69)μg/g and(50.69±15.92)μg respectively,which were close to the peak value and accounted for about 14% of the administration dosage.At 2 d,the quantity of etimicin sulfate in the kidneys of the rats began to decrease,by 50% at 4 d and 85% at 7d.At 10 d,etimicin sulfate in the tested animals was not detecsed except for one rat.Conclusion Etimicin sulfate is significantly accumulated in the kidneys of the rats after a single-dosage intramuscular injection.
