摘要
应用毛细流自组装成环荧光显微成像技术,建立了铝敏化依诺沙星的测定方法,并实测了不同样品中依诺沙星的浓度。在NH3-NH4Cl缓冲介质(pH9.7)和聚乙烯醇(PVA-124)存在下,Al3+-ENX复合物液滴在疏水性玻璃表面上受热挥发形成直径约为1.63mm,环线宽约为50μm的自组装环。当点样体积为0.20μL,其线性范围1.00×10-13~5.00×10-13mol/ring(5.00×10-7~2.50×10-6mol/L),检出限为7.65×10-15mol/ring(3.83×10-8mol/L)。本方法用于葡萄糖酸依诺沙星注射液中依诺沙星含量和牛奶中依诺沙星残留量的检测,回收率分别为96.9%~107.0%和94.2%~102.5%,相对标准偏差(RSD)分别小于4.1%和2.4%。实测了健康志愿者服用依诺沙星胶囊后尿液中依诺沙星的平均浓度及兔子灌喂依诺沙星后血清中药物平均浓度,均得到了较满意的结果。
The self-ordered ring (SOR) fluorescent microscopic imaging technique has been applied to the determination of the concentration of aluminum-sensitized enoxacin (ENX) in different samples based on the capillary effect of solvent on solid supports.In the presence of NH3-NH4Cl buffer solution (pH 9.70) and PVA-124,the droplet of Al3+-ENX chelate can form a SOR on solid supports with the diameter of 1.63 mm and its ring belt width of 50 μm.The quantitative analysis of enoxacin is achieved with the linear range of 1.00×10^-13-5.00×10^-13mol per ring(5.00×10^-7-2.50×10^-6 mol/L) and limit of detection 7.65×10^-15mol per ring(3.83×10^-8 mol/L) when 0.20 μL droplet was spotted.The technique has been applied to the determination of enoxacin in the enoxacin gluconate injection and residue in four different milk samples with the recoveries of 96.9%-107.0% and 94.2%-102.5%,RSD less than 4.1% and 2.4%,respectively.The enoxacin average concentration in healthy volunteer′s urine and rabbit serum after oral administration was satisfactorily determined with satisfactory result.
