摘要
目的建立同时测定中药独活中蛇床子素和二氢欧山芹醇当归酸酯含量的HPLC方法。方法药材样品加乙醚用索氏提取6h,水浴挥干,甲醇定容;采用MerckC18柱(250mm×4.6mm,5μm),流动相为甲醇-0.1mol/L硫酸铵溶液(70:30),用硫酸溶液(1→5)调节pH为3.0,流速为1.0mL/min,检测波长为322nm。结果理论板数以蛇床子素和二氢欧山芹醇当归酸酯计算均不低于5000,两物质的色谱峰分离度>1.5,对称因子均在0.95~1.05。蛇床子素和二氢欧山芹醇当归酸酯的线性范围分别为20.2~161.9和5.1~41.2mol/L,平均回收率为99.2%和98.8%,RSD为1.56%和2.16%。结论本实验建立的色谱方法敏感、准确、重现性好,可用于中药材独活的质量控制。
Objective To establish a method for determination of osthole and columbianctim inAngelica pubeseens. Methods To extract sample by Soxhlet extractor with ethylether. The HPLC method was chromatographed on Merck ODS C18 column(250mm×4.6mm, 5μm), the mobile phase was methanol- 0.1mol/L ammonium sulfate solution(70 : 30), as justed its acidity to pH3.0. Flow rate was 1.0mL/min, the detection was set at 322 nm. Results The calibration curve was linear in the range of 20.2-161.9mg/L(r=0.9998)for osthole and 5.1-41.2mg/L(r=0.9996)for columbianctim. The average recovery in sample was 99.2% and 98.8% for osthole(RSD 1.56%) and columbianctim(RSD 2.16%), respectively. Conelusinn The method is sensitive, accurate and simple, and it's suitable for the quality control of Angelica pubescens.
