摘要
目的:研究电针"内关"穴预处理对缺血再灌注损伤大鼠心肌的保护作用及其对心脏肥大细胞的影响。方法:18只雄性SD大鼠随机分为假手术组、模型组及电针组。电针组电针双侧"内关"穴30min。分别于左冠状动脉前降支阻断前(T0)、针刺30min后(T1)、阻断30min(T2)、心肌再灌注30min(T3)及再灌注120min(T4)检测血清肿瘤坏死因子(TNF-α)及组胺的含量,于T0、T3、T4检测血清乳酸脱氢酶(LDH)及肌酸激酶(CK-MB)的含量,于再灌注末测定心肌梗死面积、心肌组织中TNF-α和组胺的含量及肥大细胞脱颗粒百分率。结果:与假手术组比较,模型组T3、T4时血清中LDH及CK-MB含量均升高(P<0.05);与模型组相比,电针组T3及T4时间点的LDH及CK-MB含量均降低(P<0.05)。电针组的心肌梗死面积小于模型组(P<0.05)。模型组在T2、T3时血清TNF-α和组胺含量高于假手术组,电针组T2及T3时间点的血清TNF-α及组胺明显高于模型组(P<0.05)。模型组心肌组织中TNF-α及组胺均低于假手术组(P<0.05),电针组心肌组织中TNF-α及组胺含量低于模型组(P<0.05);肥大细胞脱颗粒率模型组明显高于假手术组,电针组明显高于模型组(P<0.05)。结论:电针"内关"穴预处理促进心肌肥大细胞脱颗粒,从而起到心肌保护作用。
Objective To investigate the effects of "Neiguan" (PC 6)-electroacupunture (EA) preconditioning on the myocardium and its mast cells in myocardial ischemia/reperfusion (MI/R) rats. Methods Eighteen male SD rats were randomly assigned to sham group, model (IR) group and EA group (n =6/group). MI/R model was established by occlusion of the descending anterior branch of the coronary artery. Blood samples were taken from the femoral vein before MI (To), EA for 30 min (T1), 30 min after MI (T2), 30 min after MI/R (T3) and 120 min after MI/R (T4) for assaying serum tumor necrosis factor (TNF)-α and histamine contents by using ELISA. Serum lactate dehydrogenase (LDH) and creatinkinase isoenzyme (CK-MB) le- vels were measured at T0 ,T3 and T4 by using an automatic biochemistry analyzer. The infarct size was detected by Evan's blue and tetrazolium chloride (TTO) staining. Myocardial TNF-α and histamine contents were detected by ELISA. The percentage of mast cell degranulatien was determined by toluidine blue staining. Results Following MI/R, serum LDH and CK-MB levels at phase T3 and T4, serum TNF-α and histamine contents at phase T2 and T3, and myocardial mast cell degranulation rate increased significantly, and myocardial TNF-α and histamine contents decreased in model group in comparison with pre-MI/R (P〈0.05). Compared with IR model group, serum LDH and OK-ME3 levels at phase T3 and T4, myocardial TNF-α and histamine contents all decreased significantly (P-〈0.05), but serum TNF-α and histamine contents increased significantly in EA group(P〈0.05). The infarct size was remarkably smaller in EA group than that in IR model group(P^0.05). Conclusion "Neiguan" (PC 6)-EA preconditioning has a cardioprotective effect on the ischemia-reperfusion myocardium by promoting mast cell degranulation.
