摘要 目的:初步探讨β-连环蛋白高表达对成纤维细胞趋化及上皮型钙黏连素表达的影响。方法:利用cDNA文库构建人截短β-连环蛋白表达质粒,重组质粒瞬时转染人胚肺成纤维细胞,检测成纤维细胞趋化性变化,观察上皮型钙黏连素表达情况。结果:成纤维细胞转染重组质粒后β-连环蛋白转录和翻译水平升高(25806.3±2170.1vs8216.7±833.1,P<0.05;70496.0±1960.0vs25867.3±136.5,P<0.05)。β-连环蛋白高表达促进成纤维细胞趋化(340±38vs80±14,P<0.05)。转染重组质粒后成纤维细胞表达上皮型钙黏连素减少(25806.3±2170.1vs8216.7±833.1,P<0.05;89786.3±276.7vs31876.0±1411.3,P<0.05)。成纤维细胞趋化与上皮型钙黏连素水平存在明显负相关(r=-0.988,P<0.05)。结论:β-连环蛋白高表达可促进成纤维细胞趋化,造成上皮型钙黏连素表达减少。 AIM : To investigate the effect of over - expressing β - catenin on cell chemiotaxis and epithelial cadherin expression in human embryonic pulmonary fibroblast. METHODS : Truncated human β - catenin expression plasmid was constructed using polymerase chain reaction from cDNA library. Recombinant plasmids were transfected into human embryonic pulmonary fibroblast. Cell chemiotaxis was evaluated by Transwell assay. Epithelial cadherin expression was semi - quantified after transient transfection. RESULTS: Transcription and translation of β - catenin was elevated after transfection ( 25 806. 3 ± 2170 vs 8216. 7± 833.1, P 〈0.05 ;70496.0±1960.0 vs 25 867. 3±136. 5,P 〈0.05 ) . Cell chemiotaxis increased after transfection (340±38 vs 80 ±14, P 〈 0. 05 ). Epithelial eadherin expression was semi - quantified. Over - expression of β - catenin down - regulated epithelial eadherin expression at mRNA and protein level (25 806. 3 ±2 170. 1 vs 8 216. 7 ±833.1, P 〈0.05 ; 89 786. 3 ±276.7 vs 82 026. 7 ± 1 515.7, P 〈0.05). Fibroblast chemiotaxis was negatively related to epithelial cadherin expression ( r = - 0. 988, P 〈 0. 05 ). CONCLUSION: Over - expression β - catenin in human embryonic pulmonary fibroblast promoted cell chemiotaxis and decreased epithelial cadherin expression.
