摘要
讨论了荧光法测定微生物TGase的条件,对反应体系中MDC的浓度、反应时间、DTT的浓度、硫铵的浓度、酶浓度等因素与体系荧光强度的关系进行了探讨,得出30μmol/LMDC1.0mL,2.29mg/mLDMC1.0mL,5.5mmol/LDTT0.3mL,加入一定量酶样品,37℃反应30min后用1mol/L硫铵溶液01mL灭活,测定体系在360nm激发光下,530nm处的荧光强度值,以此作为定量的依据。
An important assay method has been discussed for microbial transglutaminase regarding dansyl cadaverine incorporation into casein.It is known that the amine,after incorporation into protein by transglutaminase,shows a marked increase of fluorescence accompanied by a slight blue shift.The parameter such as:the concertration of MDC,the reaction time,the concertration of DTT and ammonium sulfate were studied and got better results.The results is:30 μmol/L MDC 1.0 mL,2,29 mg/mL DMC1.0 mL,5.5 mmol/L DTT0.3 mL,37 ℃,30 min.
