摘要
本实验以茉莉花茶为供试材料,优化茶多糖的提取工艺,旨在提高茶多糖的提取率,为茶叶深加工提供理论依据。在单因素实验的基础上,利用Box-Benhnken的中心组合设计,选定温度、水料比和沉淀茶多糖时所用的乙醇浓度3个因素分别选3水平进行中心组合实验,通过响应面分析实验,拟合出数学模型:Y=6.55+1.30A+0.83B+1.10C-0.092AB+0.11AC+0.34BC-1.56A2-0.64B2-1.48C2。利用该函数关系来优化茶多糖提取条件,最终确定茶多糖的最佳浸提条件为:浸提温度100℃;水料比为26.8;乙醇浓度为90%。在该条件下茶多糖得量有所提高,且验证值为7.8610mg/g,比单因素最高提取率高25.6%,表明响应面法可有效用于茶多糖提取方法的优化。
In order to increase the extraction yield of tea polysaccharidem and provide theory evidence for process of coarse tea,the extraction technology of tea polysaccharides of jasmine tea bought from market was investigated in this paper.On the basis of single-factor experiments,the Box-Behnken center-united experimental design principles were used to design the experiments and the response surfaces analysis of 3 factors,including extracting temperature,water to material ration and ethanol concentration when deposited tea polysaccharides,and 3 levels were adopted respectively.Obtained by responsive surfaces analysis and fitted the mathematical model which was Y=6.55+1.30A+0.83B+1.10C-0.092AB+0.11AC+0.34BC-1.56A2-0.64B2-1.48C2,the optimum combination was 100℃ extracting temperature,26.8 water to material ration and 90% ethanol concentration,under which the yield of TPS highly reached with 7.861 0 mg/g,TPS yield after optimization is 25.6% higher than the best yield in single factor experiment,which indicated that the response surface analysis was effective for optimization of tea polysaccharides extraction technology.
