摘要
为了获取高质量的卵泡卵母细胞,提高绒山羊体外胚胎生产的效率,采用切割法与抽吸法采集卵泡卵母细胞,并对获得的可用卵母细胞进行了体外成熟培养,表明结果:切割法获得的可用卵母细胞较多,明显增加了用于体外成熟的卵母细胞数。在温度37.0、38.0、39.0℃条件下对卵母细胞进行体外成熟培养,成熟率分别为67.74%、72.00%、75.38%,表明39.0℃适合辽宁绒山羊卵母细胞体外成熟培养。在温度39.0℃条件下用10%胎牛血清(FCS)的TCM-199培养液培养卵母细胞的成熟率为78.57%,而在10g/LBSA的TCM-199培养液中培养卵母细胞的成熟率为74.07%,二者差异不显著。
In order to gain the follicular oocytes with high quality and to improve the frequency of in vitro production of cashmere goat embryos,the dissection method and the aspiration method were applied to collect the follicular oocytes of the goats,and in vitro maturation of the available oocytes was performed. Results showed that the available oocytes obtained by the dissection method were more than those by the aspiration method,and the number of oocytes suitable to be used in vitro maturation obviously increased. The cashmere goat oocytes were cultured at 37.0,38.5,and 39.0 degrees C,and the maturation rates were 67.74%,72.00%,and 75.38% respectively. The maturation rate of oocytes cultivated in TCM-199 culture medium containing 10% FCS at 39 degrees C reached 78.57%,while that of oocytes in TCM-199 culture medium containing 10 g/L bovine serum albumin was 74.07%.
