摘要
目的探讨不同分化程度的鼻咽癌细胞株X线辐射后miR-7表达的变化。方法 2种细胞均分为对照组(未辐射)、2Gy组、6Gy组和10Gy组,X线辐射细胞株,线性二次模型拟合CNE-1、CNE-2细胞存活曲线,Trizol法提取细胞总RNA。茎环法特异性逆转录miR-7,染料法实时定量PCR,以对照组CNE-1细胞为参照样本,ΔΔCt法得出各样品miR-7的相对倍数。结果对照组CNE-2细胞miR-7的量是CNE-1的2.51倍。2Gy组CNE-1细胞miR-7表达量增加,CNE-2细胞则由早期的49.44下降到晚期的0.15。6Gy组两种细胞miR-7含量差别不大。10Gy组CNE-1的miR-7含量高于CNE-2。结论分化程度不同的鼻咽癌细胞经不同剂量的X线辐射后,miR-7表达量各不相同。分化程度不同的鼻咽癌细胞的放射敏感性差异,可能是由调控miR-7表达能力不同造成的。
【Objective】To investigate the expression patterns of miR-7 in variously differentiated nasopharynx cancer cells after radiation. 【Methods】Cells were divided into four groups: control group, 2Gy group, 6Gy group and 10 Gy group. Nasopharynx cancer cell lines CNE1 and CNE2 were irradiated by different dose of X-ray. The cell survival curves were fitted with linear-quadratic model. Total RNAs of cell lines were extracted by Trizol in 2 hours and 10 hours after radiation. The reverse transcription of miR-7 was done by Stem-loop primer. Non-irradiated CNE1 cell was reference sample and relative quantity of other samples was counted by ΔΔCt method after real time PCR used SYBR Green. 【Results】Expression level of miR-7 in CNE-2 was 2.51 times higher than that in CNE-1 of control group. In 2 Gy group expression of MiR-7 in CNE-1 increased; in CNE-2, in earlier period, miR-7 increased to 49.44 and then decreased to 0.15 in later period. There was not distinguishing expression of miR-7 in two cell lines in 6 Gy group. In 10 Gy group, CNE-1 expressed more miR-7 than CNE-2.【Conclusions】 The regulation of miR-7 expression may be the reason of different radiosensitivity of variously differentiated na-sopharynx cancer cell.
