摘要
探索了蓝隐藻的实验室自行培养条件;改进了完整类囊体膜的制备方法,获得了状态较为理想的类囊体膜,其Chla/c比值可稳定在4左右.此外,建立并优化了色素-蛋白复合物的增溶以及蔗糖密度梯度离心分离的条件,得到了4种活性的色素-蛋白复合物,经光氧化活性测定,初步判定为捕光复合物Ⅰ、捕光复合物Ⅱ以及PSⅡ颗粒和PSⅠ颗粒.实验结果为今后进一步研究蓝隐藻光合系统结构奠定了基础.
The cultural conditions of Chroomonas placiodea in laboratory are investigated,and the preparation of intact thylakoid is improved by which thylakoids in well conditions are obtained with Chl a/c ratios steady around 4.0.Moreover,the method of detergent resolution, along with the isolation by sucrose gradient centrifugation of pigment-protein complexes are established and optimized.4 active pigment-protein complexes are resolved which are designated as light-harvest complex I,light-harvest complex II,PS I and PS II pellets respectively according to the results of photooxidative activity assay.The present work will lay a solid foundation for further structural study of photosystems of C.placiodea.
