未成熟日本血吸虫卵的组分蛋白及其免疫学特性的研究

摘要 应用SDS-PAGE对日本血吸虫4周、5周和6周-SEA进行分析,分别出现21、14和20条组分蛋白带,三者间既有共同组分又存在差异,显示出不同的分子基础。于攻击感染后0~8周作动态观察,ELIB结果表明,4周-SEA及其免疫鼠血清的免疫学特性与5周-SEA和6周-SEA存在一定差异,其中被4周-SEA免疫鼠血清识别的72.4~68.5ku蛋白宽带至攻击感染后6~8周消失,与该抗原免疫鼠肝内虫卵数减少、虫卵发育延缓及肉芽肿缩小(在6周前)及后来的恢复(8周)基本一致。 The 4wk-, 5wk-and 6wk-SEA of Schistosoma japonicum were separated by sodium dodecyl sulfate dodecyl sulfate polyacrylamide gelelectrophoresis(SDS-PAGE)and 21, 14 and 20 protein bands were demonstrated respectively, not only including some common components but also some different ones,shouing the specific molecular basis of the 3 stage egg antigens.Then a kinetic observation on the pattems of recognized epitopes of the 3 stage egg antigens was dried out during 0~8 wks after chllenge infection. The enzyme linked immune blotting (ELIB)analysis of 4wk-SEA,5wk-SEA and 6wk-SEA showed some difference in the immunological characterisitics,and changeable epitopes recognized by the immunized sera. Interestingly,the 73~69 kDa bands, which were recognized by the 4wk-SEA mediated sera, were not seen at 6~8 wks after challenge infection and this finding would be associated with the reduction of liver egg number and inhibition of egg development as well as the decreased pouloma size in immunized mice before 6wks.
