摘要
目的探讨银杏提取物EGB761对过氧化氢诱导肾小管上皮细胞凋亡的抑制作用及发生机制。方法采用500μmol/L浓度的过氧化氢(H2O2)作用于对数生长期的肾小管上皮细胞(NRK52E),建立细胞氧化损伤模型。银杏提取物EGB761分为高中低3个浓度组进行干预,应用细胞生长曲线检测细胞生长情况,用流式细胞术检测肾小管上皮细胞的凋亡率,RT-PCR检测Caspase-3基因的表达差异。结果与过氧化氢组比较,高、中浓度组银杏提取物EGB761的凋亡阳性率均明显降低,而低浓度组虽有减少趋势但无显著性差异;高、中浓度组银杏提取物EGB761的Caspase-3表达阳性率较过氧化氢组明显降低,且随着浓度的增加而表达逐渐减少(P<0.05)。结论银杏提取物EGB761可以拮抗过氧化氢诱导肾小管上皮细胞凋亡的效应,其机制估计与下调Caspase-3的表达有关。
Objective It is to explore the inhibiting effects of Gingko extraction EGB761 against renal tubular epithelial cell apoptosis induced by H2O2 and its mechanism.Methods NRK-52E cells were treated with 500 μmol/L H2O2 to made oxidative stress-induced injury models.The EGB761 was divided into 3 groups of high,mild and low dosage to prevent cell apoptosis.The cell proliferation was evaluated by MTT assay.Apoptosis rate was assessed by flow cytometry.The expression of Caspase-3 was detected by RT-PCR.Results Compared with H2O2 group,the apoptosis rates of high and mild dosage EGB761 groups decreased obviously,but that in low dosage group although decreased,but there was no significant difference.The positive expression rate of Caspase-3 in high and mild dosage EGB761 groups was obviously lower than that in H2O2 group,and the expression decreased more with the concentration increased(P〈0.05).Conclusion Gingko extraction EGB761 can inhibit the renal tubular epithelial cell apoptosis induced by H2O2,its mechanism may be that it can decrease the expression of Caspase-3.
