摘要
目的研究海洋刺参多糖(SJAMP)对人宫颈癌细胞系HeLa细胞周期影响并探讨其作用机制。方法实验分为对照组(不用SJAMP)及1.6、3.26、.4 g/L不同剂量SJAMP组。应用免疫细胞化学法分别检测各组SJAMP对增殖细胞核抗原(PCNA)、细胞周期抑制蛋白Mdm2表达的影响;流式细胞仪检测不同浓度SJAMP对HeLa细胞周期的影响。结果与对照组相比较,1.6、3.2和6.4 g/L SJAMP组均能抑制PCNA和Mdm2的表达(F=209.02~951.11,q=18.97~74.55,P〈0.05),且具有一定的浓度依赖和时间依赖关系。经SJAMP处理的HeLa细胞发生G1期阻滞,对照组及1.6、3.2、6.4 g/L SJAMP组细胞的凋亡率分别为0.25%、4.32%、11.36%和30.04%,差异有显著性(F=26.95~259.21,q=3.34~36.36,P〈0.05)。结论 SJAMP对HeLa细胞的生长具有明显的抑制作用。其机制可能是SJAMP降低PCNA和Mdm2蛋白的异常表达,使细胞周期发生G1期阻滞,从而起到抑制细胞增殖的作用。
Objective To investigate the effect of Stichopus japonicus acidic mucopolysaccharide(SJAMP) on cell cycle of HeLa cells and its mechanisms. Methods HeLa cells were exposed to various concentrations of SJAMP for certain time.Immunocytochemical stain was used to detect the expressions of PCNA and Mdm2 protein.Flow cytometric analysis was performed for cell cycle progression. Results A dose-dependent as well as time-dependent growth inhibition was observed in SJAMP-treated HeLa cells,and associated with an increase in G1 arrest,the rate of APO was 0.25%,4.32%,11.36% and 30.04%,respectively;different concentrations of SJAMP decreased the abnormal expression of Mdm2 gene and strongly decreased the expression ofPCNA(F=209.02-951.11,q=18.97-74.55,P0.05). Conclusion SJAMP has an inhibitory action on proliferation of HeLa cells and its mechanism might be related to lowering abnormal expressions of PCNA and Mdm2,blocking cell cycle,resulting in the inhibition of cell multiplication.
