摘要
研究了碱性蛋白酶分别与木瓜蛋白酶、中性蛋白酶、胃蛋白酶组合对β-乳球蛋白进行双酶解,以水解度(DH)和可溶性氮含量(TCA-SN)表征其反应程度,确定碱性蛋白酶与木瓜蛋白酶为最佳酶解组合。通过正交实验,确定其组合最佳反应条件为:β-乳球蛋白先径酶活添加量为5310U/g的碱性蛋白酶水解150min后,再经酶活加量为4500U/g的木瓜蛋白酶水解,反应到180min时,水解度可达36.21%。
Alkaline protease was combined individually with papain, neutral protease, pepsin to hydrolyzed β- laetoglobulin. Degree of hydrolysis (DH) and TCA-SN was indexed to indieate reaction degree. The results showed that the best combination is alkaline protease with papain. The optimum parameters of the dual-enzyme hydrolysis were: Firstly, alkaline protease was hydrolyzed for 150min where enzyme amount of alkaline protease 5310U/g; then added papain where enzyme amount papain was 4 500U/g till 180min, and the hydrolysis degree reached 36.21% .
