摘要
目的:建立一种简单实用的胶体金免疫层析法用于食品样品中大肠杆菌O157:H7的快速筛查。方法:采用柠檬酸三纳还原法制备胶体金颗粒.标记抗E.coli0157:H7单克隆抗体.制备免疫胶体金复合物,组装胶体金免疫层析快速检测试纸条,并对其特异性、灵敏度和稳定性进行了测试和评价。结果:该试纸条特异性较好.不与其他受试菌株发生交叉反应,检测灵敏度为10^5CFU/mL。120份人工污染E.coliO157:H7模拟食品样本增菌后检测,该法检测阳性118份,灵敏度高达98.3%。结论:本实验成功研制了E.coliO157:H7胶体金免疫层析试纸条,该试纸条具有灵敏度高、特异性好、检测速度快、操作简便等优点,可用于现场大量样品的初筛。
Objective To establish a simple, practical gold immunochromatography assay (GICA) for rapidly screening E.coli O157:H7 in food samples. Methods Trisodium citrate reduction was used to prepare colloidal gold pellets, which were coupled with monoclonal anli-E.coli O157:H7 to produce immune complex and assemble a GICA strip tool. The sensitivity and specificity of this strip was then evaluated. Results The GICA strip exhibited a higher specificity for detecting E.coli O157:H7 and did not cross-react with other strains. The sensitivity of the strip was l0s cfu/ml of pure bacterial culture. 118 of 120 samples artificially contaminated with E.coli O157:H7 were detected positive, with a sensitivity of up to 98.3%. Conclusions A GICA strip for detecing E.coli O157:H7, which has a higher specificity and a better specificity, is successfully produced. It is a rapid, simple method for detection and can be used for primarily screening food samples on site.
