摘要
目的探讨茜草醇提浸膏对刀豆蛋白A(ConA)致小鼠急性肝损伤的保护作用。方法采用昆明种小鼠,灌胃给予6、10、16g.kg-1.d-1的茜草醇提浸膏和联苯双酯滴丸溶液(0.2g.kg-1.d-1)灌胃。14d后尾静脉注射ConA20mg.kg-1制备小鼠急性肝损伤模型,8h后取血用化学比色法检测血清ALT、AST活性;同时用酶联免疫法测定肝组织匀浆中IL-1β、TNF-α、IL-4、IL-10含量的变化及观察肝脏病理学变化。结果与模型对照组相比,茜草醇提浸膏16g.kg-1.d-1组小鼠ALT、AST活性明显降低(P<0.01),肝组织匀浆中IL-1β、TNF-α的含量明显下降(P<0.01),IL-4、IL-10含量升高(P<0.01),能明显改善肝组织的病理学损伤。结论茜草醇提浸膏对ConA所致小鼠急性肝损伤具有保护作用。
Objective To study the protective effect of ethanol extracts from Radix Rubiae Cordifoliae on hepatic injury induced by ConA. Methods 60 mice of Kunming species were randomizedly allocated into 6 groups. Alcohol extraction from Radix Rubiae Cordifoliae with different doses (6,10,16g · kg-1 ·d- 1 ) combined with bifendate diester drop pills(0.2g · kg-1 · d-1) were administrated into gaster. After 14 days, the mice were induced to hepatic injure by subcutaneous injection by ConA (20mg · kg-1) in tail vein. Then we detected the levels of ALT and AST in plasma and levels of IL - 1, TNF - α, IL - 4 and IL - 10 in hepatic ho- mogenate, and observed the pathological change in liver. Results Compared with the control group, the levels of ALT , AST , IL - 1 β and TNF - α in mice were significantly reduced and the levels of IL - 4 and IL - 10 were significantly increased in the mice with the oral administration of alcohol extracts from Radix Rubiae Cordifoliae ( all P 〈 0.01 ). Conclusion Alcohol extracts from Radix Rubiae Cordifoliae can take a protective role in hepatic injury induced by ConA in mice.
