摘要
目的探讨合肥地区汉族人群G蛋白β3亚单位基因C825T多态性(GNβ3C825T)与2型糖尿病(T2DM)合并大血管病变(MA)的遗传易感性。方法采用聚合酶链反应结合限制性内切酶片段长度多态分析方法(PCR-RFLP)检测293例合肥地区汉族T2DM患者GNβ3C825T多态性,并测定T2DM患者的体质指数(BMI)、腰臀比(WHR)、收缩压(SBP)、舒张压(DBP)、空腹血糖(FBG)、餐后2 h血糖(P2hPG)、胰岛素敏感指数(ISI)等指标。结果 T2DM无MA组GNβ3C825T中基因型频率(CC为30.6%,CT为50.0%,TT为19.4%)、等位基因频率(C为55.6%,T为44.4%)与T2DM有MA组基因型频率(CC为29.3%,CT为48.8%,TT为21.9%)、等位基因频率(C为53.7%,T为46.3%)分布差异无统计学意义,在调整年龄、性别、吸烟、高血脂、高血压后,Logistic回归分析显示在糖尿病人群中MA与GNβ3C825T无关。结论合肥地区汉族人群中GNβ3C825T多态性可能与T2DM合并MA遗传易感性无关。
Objective To explore the heredity susceptivity between C825T polymorphism of human G-proteinβ3 subunit gene in the Chinese Han population with type 2 diabetes mellitus(T2DM) complicated by macro-vascular(MA) disease in Hefei area.Methods Polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) was used to test the polymorphism of GNβ3C825T of 293 T2DM patients(41 case with MA).AT the same time,record T2DM patients' body mass index(BMI),waist-hip ration(WHR),systolic blood pressure(SBP),diastole blood pressure(DBP),fasting blood glucose(FBG),blood glucose of postprandial 2 hours(2hPG) and insulin sensitive index(ISI).Results The genotype frequency and allele frequency of T2DM without MA group were respectively CC/CT/TT=30.6%/50.0%/19.4%,C/T= 55.6%/44.4%;while the genotype frequency and allele frequency of T2DM with MA group were respectively CC/CT/TT=29.3%/48.8.0%/21.9%,C/T=53.7%/46.3%.there were no significant difference frequencies in genotype and allele of GNβ3 between two groups.After adjusting factors such as age,gender,smoking,hyperlip,and hypertension,stepwise multivariate unconditional logistic regression analysis showed the GNβ3C825T polymorphism was not related to T2DM with MA.Conclusion The GNβ3C825T polymorphism has no heredity susceptivity to MA among T2DM patients in the Chinese Han population of Hefei region.
