摘要
目的:建立并采用气虚冻伤大鼠模型,从血流变和TXB2、6-k-PGF1α方面探查黄芪桂枝五物汤对气虚冻伤模型大鼠的生物效应及作用机理。方法:30只Wistar大鼠随机分为正常对照组、气虚冻伤模型组、黄芪桂枝五物汤组3组,每组10只。采用气虚模型(过度疲劳+饮食失节法)复加冻伤模型(低温乙醇和水混合浸泡法)制备大鼠气虚冻伤模型。造模后第2天,黄芪桂枝五物汤组按相当于生药材4.7 g/(kg.d)体重给予灌胃,正常对照组和气虚冻伤模型组给予等量蒸馏水,每日2次。连续给药3 d后,测定各组大鼠血流变、TXB2、6-k-PGF1α的变化。结果:造模后第3天,与空白对照组比较,气虚冻伤模型组全血黏度、红细胞聚集指数、TXB2显著升高;红细胞变形指数、6-k-PGF1α显著降低;与气虚冻伤模型组比较,黄芪桂枝五物汤组全血黏度、血浆黏度、红细胞聚集指数、TXB2显著降低;红细胞变形指数、6-k-PGF1α显著升高。结论:本气虚冻伤模型造模3 d存在血栓易形成状态,黄芪桂枝五物汤对气虚冻伤有一定的作用,其机理可能为通过对血循环的影响而达到益气活血疗伤的目的。
Objective:To explore the effects and mechanism of Huangqi Guizhi Wuwu Decoction(HGWD)on hemorheolo-gy,contents of TXB2 and 6-k-PGF1α of qi deficiency combined with frostbite model rats.Methods:30 Wistar rats were randomly divided into normal control group,qi deficiency combined with frostbite model group and HGWD group.Model rats were constructed by overtiredness and irregular diet combined with soaking feet in hypothermia ethanol-water mixture.3 days after treatment,hemorheology,contents of serum TXB2 and 6-k-PGF1α were determined by enzyme-linked im-munosorbent assay method.Results:Compared with normal control group,blood viscosity,plasma viscosity,erythrocyte ag-gregation index,TXB2 level in model group rats were significantly increased,erythrocyte deformability index and 6-k-PGF1α level were significantly decreased.Compared with model group,blood viscosity,plasma viscosity,erythrocyte aggre-gation,TXB2 level in HGWD group rats were significantly decreased,erythrocyte deformability index and 6-k-PGF1α level were significantly increased.Conclusion:Qi deficiency combined with frostbite model rats has a tendency to thrombosis.HGWD has therapeutic effect on qi deficiency combined with frostbite model rats through improving blood circulation and regulate the balance between TXB2 and 6-k-PGF1α.
