摘要
目的:观察蝎毒抗癌多肽(antineoplasticpolypeptidefromButhusmartensivenom,APBMV)对人低分化鼻咽癌细胞(CNE2Z)的毒性作用,从而找出APBMV抑制人鼻咽癌的最佳浓度。方法:用MTT法测定APBMV对人低分化鼻咽癌细胞(CNE2Z)的毒性。CNE2Z细胞培养于RPMI1640培养液中;选增生活跃,形态良好的细胞接种在96孔培养板。细胞浓度为5×104ml-1。细胞被分为10个实验组,MMC阳性对照组、阴性对照组和8个APBMV实验组,实验组的APBMV质量浓度分别为1mg·L-1,2mg·L-1,3mg·L-1,4mg·L-1,5mg·L-1,6mg·L-1,7mg·L-1,8mg·L-1。加药后,细胞培养48h,然后加入MTT10μl·孔-1,继续培养4h,加入200g/LSDS500g/LDMF混合溶液溶解细胞内的结晶体,测定560nm和630nm波长处的D值,并计算生长抑制率(Growthinhibitingrate,GI)。结果:APBMV在2~8mg·L-1浓度范围内,GI<50%,CT>50%,其中在3~6mg·L-1浓度范围内,CT?
Aim:To determine cytotoxic effect of antineoplastic polypeptide from Buthus
Martensii venom (APBMV)on human poor differential nasopharyngeal carcinoma cells(CNE
2Z),and find out the suitable concentration of APBMV for inhibiting CNE 2Z Methods[ :By
using MTT method cytotoxic effect of APBMV on CNE 2Z cells were investigated. CNE 2Z cells
were cultured in RPMI 1640 medium. Actively proliferating CNE 2Z cells were inoculated to 96
well culture plate at concentration of 5×10 4 cell· ml -1 . The cells were divided into 10
groups:MMC positive contral group,negative control group and 8 experimental groups. APBMV
at concentration of 1 mg·L -1 to 8 mg·L -1 were added to cells of 8
experimental groups seperately.After the cells were cultured for 48 h, MTT 10 μl/well were
added. The cells were then cultured for another 4 h, and 200 g/L SDS 500 g/L DMF mixing
solution was used to dissolve the crystals in the cells. OD value in 560 nm and 630 nm
wave was detected and growth inhibiting rate (GI) of cells was calculated. Results: The
final concentration of APBMV was in the range of 2~8 mg·L -1 , GI of cells was less than
50%and cytotoxicity (CT) was higher than 50%. Cytotoxicity increased higher than 68%when
the final concentrations of APBMV was in the range of 3~6 mg·L -1 . The differences
were very significant ( P <0.05 or P <0.01) compared with control.But the dose response
relationship was not very obvious.Conclusion:APBMV possesses the cytotoxic effect on CNE
2Z cells.
