摘要
目的:研究鱼腥草素β环糊精包合物的制备方法及包合前后鱼腥草素物理性能变化。方法:采用研磨法制备鱼腥草素β环糊精包合物,含量测定方法是将鱼腥草素与铜(Ⅱ)于氨性介质中形成鳌合物,于(300±1)nm波长处测定螯合物氯仿提取液吸光度,测定了包合物及鱼腥草素的溶解度、溶出速度,比较了二者X射线衍射图谱、紫外吸收光谱变化及其在水中溶解稳定性。结果:鱼腥草素β环糊精包合物的主客分子比为1∶1,包合后,鱼腥草素的溶解度增大了11.4倍,鱼腥草素及包合物的溶出速度参数T50%分别为1.7,1.0min,Td分别为3.0,1.4min,二者紫外吸收光谱完全一致,包合后鱼腥草素的晶体衍射峰消失。结论:鱼腥草素与β环糊精形成了新的物相,包合后,鱼腥草素溶解度、溶出速度及稳定性增强,并可掩盖其不良气味。
OBJECTIVE:To study the preparation method of the inclusion complex of Decanoyl acetaldehyde(DA) β cyclodextrin( β CD) and the changes of the physical properties of DA before and after being included by β CD.METHOD:The inclusion complex of DA β CD was prepared by milling method.The quantitative method of DA was based upon the formation of chelate compound from DA and copper(Ⅱ) in ammoniacal medium.The chelate was extracted into chloroform and the absorbance of the solution was measured at 300±1nm against a chloroform blank.The solution and dissolution rates of inclusion complex and DA were measured,It were compared the changes of X ray diffractin and UV spectrum,and the stability in water between DA and inclusion complex.RESULTS:The molecular ratio of DA to β CD in the inclusion complex was 1∶1.The solubility of DA in inclusion complex was 11.4times higher than that of pure DA.The parameters of dissolution rate of DA and inclusion complex T 50% were 1.7 and 1.1 min respectively,Td were 3.0 and 1.4min respectively.Their UV spectrum of DA showed no difference.The diffraction peak of DA crystal disappeared after being included by β CD.CONCLUSION:A new physical phase forms between DA and β CD.the solubility、dissolution rate and stability in water of DA can be increased,and the bad smell of DA can be masked after being included by β CD.
