摘要
目的:探讨胃肠舒促胃肠动力作用机制。方法:将50只Wistar大鼠随机分为正常对照组、模型对照组、胃肠舒小剂量组、胃肠舒大剂量组、西沙必利组。除正常对照组外均制备胃肠动力障碍模型。各组用等容积受试药液灌胃,连续7d,将各组动物麻醉选取胃肠标本。免疫组化法观察AchE及NO的表达变化,图象分析仪测定肌间神经丛光密度值。结果:胃肠舒能显著升高胃肠组织中AchE、NOS(P<0.01)。结论:胃肠舒促进胃肠动力作用机制:①通过NO介导加强胃肠舒张,反射性引起AchE释放增加,使二者恢复协调平衡,从而增加胃肠平滑肌细胞线粒体跨膜电位和ATP生成,改善胃肠平滑肌细胞动能而促进胃肠运动。②通过胃肠道AchE作用于平滑肌M受体,开放质膜上L型Ca2+通道,使Ca2+内流而加强胃肠平滑肌收缩。
Objective: Investigating the effect of Weichangshu on gastrointestinal motility. Methods: Divide 50 Wistar rats into 5 groups randomly:normal control, model control, low dosage of Weichangshu, high dosage of Weichangshu, Cisapride, 10 in each group.In addition to the normal control group,the other four groups were prepared animal models of gastrointestinal motility disorder;the groups were given the same volume solution of medicine by way of gastric perfusion. for seven days, samples were taken from antrum and campylobacter after animals were anesthetized.AchE,NO were showed with immunohistocheinical method by SP.The optical density of myenteric plexues were measured with mitrosperegraph. Results: groups of Weichangshu can effectively increase AchE,NOS in gastro-intestinal tractl (P0. 01).Conclusion :The mechanism of Weichangshu promoting gastrointestinal motility: ①it can reinforce the gastrointestinal relaxation mediated by NO,then increases the secretion of AchE as a reflection to keep the refined balance between NO and AchE, which increases the content of ATP and mitochondrial transmembrane-potential and changes the Kinetic energy of gastrointestinal smooth muscle cells. ②It can increase AchE,which act on M-receptor and opens L Ca2+ channel on protoplasmic membrane and cause gastrointestinal smooth muscle contraction.
