血管内皮细胞生长因子及地塞米松对早产大鼠肺发育的影响 被引量:2

Effects of Vascular Endothelial Cell Growth Factor and Dexamethasone on Lung Development of Premature Rats in Vitro

摘要 目的观察血管内皮细胞生长因子(VEGF)及地塞米松(DEX)对早产大鼠肺表面活性物质(PS)的影响,探讨VEGF及DEX与肺发育的关系。方法孕15 d清洁级Wistar大鼠12只,随机分为VEGF组、DEX组、VEGF+DEX组和9 g.L-1盐水组,每组3只。应用薄层层析法测定各组孕鼠剖宫取出的胎鼠(每组至少留6只)肺组织PS中总磷脂(TP)、磷脂酰胆碱(PC)、磷脂酰甘油(PG)、鞘磷脂(SM)水平。采用SPSS 10.0软件进行统计学分析。结果 VEGF组TP、PC、SM和PG水平分别为(69.18±2.92)μg.g-1、(42.62±1.21)μg.g-1、(17.50±0.98)μg.g-1、(6.10±0.12)μg.g-1,DEX组TP、PC、SM和PG水平分别为(84.10±6.30)μg.g-1、(47.10±4.04)μg.g-1、(22.60±1.05)μg.g-1、(5.97±0.12)μg.g-1;VEGF+DEX组TP、PC、SM和PG水平分别为(105.70±10.75)μg.g-1、(62.00±2.25)μg.g-1、(30.20±2.01)μg.g-1、(6.36±0.15)μg.g-1;9 g.L-1盐水组分别为(57.18±3.40)μg.g-1、(33.22±0.90)μg.g-1、(13.50±0.58)μg.g-1、(5.10±0.20)μg.g-1。VEGF组TP、PC、SM及PG水平与其他组比较,差异均有统计学意义(Pa<0.01),任意二组TP、PC、SM、PG水平比较,差异均有统计学意义(Pa<0.01)。结论 VEGF能促进PS的合成和分泌,改善肺功能,有利于防止因PS缺乏所致的早产鼠呼吸窘迫综合征的发生。 Objective To observe the effects of vascular endothelial cell growth factor(VEGF) and Dexamethasone(DEX) on lung development of premature Wistar rats in vitro, and explore the relationship of lung development with VEGF and DEX. Methods Wistar rats at 15 days gestation, were randomly divided into 4 groups : VEGF group, DEX group, VEGF ± DEX group and control group, 3 rats in each group. Content of total phospholipid ( TP), phosphatidylcholine ( PC ), phosphatidyl glycerol ( PG ), sphingomyelin ( SM ) were measured by thin layer chromatography. SPSS 10.0 software was used to analyze the data. Results The contents of TP, PC, SM and PG in VEGF group were (69.18±2.92) μg·g^-1,(42.62±1.21) μg·g^-1,(6.10±0.12) μg·g^-1,and (17.50±0.98)μg·g^-1;thosein DEX group were (84.10±3.30)μg·g^-1,(47.10±4.04) μg·g^-1,(22.60±1.05)μg·g^-1 and (5.97±0.12) μg·g^-1;the contents of TP,PC,SM and PG in VEGF±DEX group were (105.70±10.57)μg·g^-1,(62.00 ±2.25)μg·g^-1,(30.20 ±2.01)μg·g^-1,(6.36 ±0. 15)μg·g^-1,contents of TP, PC,SM and PG in control group were (57.18 ± 3.40)μg·g^-1, (33.22 ± 0.90)μg·g^-1, ( 13.50 ± 0.58)μg·g^-1, (5.10 ± 0.20)μg·g^-1. Content of TP, PC, SM, PG had significant differences between VEGF group, DEX group, VEGF + DEX group and control group( Pa 〈 0.01 ). Conclusions VEGF can increase pulmonary surfactant synthesis and secretion of the lung tissues in vitro,improve function of the lung tissue. It is possible to prevent the neonatal nespiratory distress syndrome occurence by absence of pulmonary smfactant.
