摘要 采用响应面法对多粘类芽孢杆菌(Paenibacillus polymyxa)产多粘菌素B的发酵条件进行了优化。在单因素实验的基础上确定了对产量影响较大的3个因素为初始pH、发酵温度和发酵时间。采用B时o间x-B37eh.7n2kehn。实在验此设条计件进下行进响行应发面酵分验析证优,实化验,确值定和了模最型佳预发测酵值条之件间为比:较初接始近pH,有6.较72高,温的度吻3合0.6度4℃,相,发对酵误差较小。多粘菌素B的产量可达到915.77mg/L,比优化前提高了33.7%。 The fermentation conditions of polymyxin B production by Paenibacillus polymyxa was optimized using response surface method. Through the single factor experiment, three factors were confirmed to have significant effect on polymyxin B production, that is, the initial pH, fermentation temperature and fermentation time. Using Box-Behnken experimental design for response surface optimization, the optimum fermentation conditions were: initial pH value of 6.72, temperature 30.64℃ , fermentation time 37.72 h. Under these conditions, the fermentation validation showed that the experimental value was comparable to model prediction, having high inosculating degree and the error was relatively small. The yield of Polymyxin B could reach 915.77 mg / L, which is 33.7% higher than before optimization.
