摘要
为探讨苯那普利对胰岛素敏感性的影响,我们以高胰岛素诱导培养法复制的胰岛素抵抗(IR)原代大鼠肝细胞模型为研究对象,采用3H一葡萄糖掺入试验等技术,了解含有苯那普利活性形式一苯那普利拉的大鼠血清对IR细胞模型及对照大鼠肝细胞的胰岛素刺激下的葡萄糖掺入率、胰岛素酶活性及胰岛素受体数目的影响。结果显示,含苯那普利拉的血清能显著提高IR细胞模型及对照细胞的胰岛素介导的葡萄糖摄取和利用能力,对IR细胞模型的作用尤为显著。但对胰岛素酶活性及胰岛素受体数目无明显影响。本研究提示,苯那普利在体内的活性形式能提高胰岛素靶细胞的胰岛素敏感性,并对皿的发展有一定的阻止作用。
To investigate the effect of benazepril on insulin sensitivity, in the insulin- resistant rat hepatocytes inducedwith high concentration of insulin and the control rat hepatocytes- the insulinstimulated glucoseincorporation rate, insulinase activity and the number of insulin receptor were determined with 3H-glucoseincorporation test and other techniques after both of them were treated with the rat serum containing the activeform of benazeprilfbenazeprilat).The results showed that the rat serum containing benazeprilat can increased theability of glucose uptake and metabolism. of the two kinds of hepatocytes, especialy the insulin-resistanthepatocytes, No obvious effect of the serum on insulin-degrading enzyme activity and the number of insulinreceptor was observed. The study suggests the active form of benazepril can improve the insulin sensitivity ofinsulin target cells and hamper the development of insulin.resistant state
