摘要
目的:研究Th17细胞及其相关细胞因子白细胞介素-17(IL-17)在胶原诱导性关节炎(collagen-induced ar-thritis,CIA)小鼠体内的变化。方法:利用Ⅱ型胶原蛋白建立小鼠CIA模型。应用流式细胞术(FCM)分析小鼠脾脏及引流淋巴结中Th17细胞的比例,采用酶联免疫吸附试验(ELISA)检测小鼠血清中IL-17的含量。结果:CIA小鼠脾脏及引流淋巴结中Th17细胞的比例分别为(3.19±0.70)%、(3.54±0.97)%,显著高于正常对照组的(0.95±0.34)%、(1.31±0.47)%,P<0.01。CIA小鼠血清中IL-17的含量也明显高于正常对照组(P<0.05)。结论:Th17细胞及其分泌的细胞因子IL-17在CIA小鼠体内明显升高,并与疾病的发生密切相关。
Objective: To study the expression of Th17 cells and their cytokines ( IL-17 ) in the development. of collagen-induced arthritis (CIA) in mice. Methods: The CIA models were immunized with collagen type Ⅱ and Freund's adjuvant. The percentage of Th17 cells in spleens and draining lymph nodes were determined by flow-cytometry, and the concentration of IL-17 in the serum were examined by enzyme linked immunosorbent assay (ELISA) respectively. Results: Compared with the normal group, CIA mice had a significantly higher percentage of Th17 cells in spleens and draining lymph nodes (P 〈 0.01 ) and had an increased concentration of IL-17 in the serum (P 〈0.05). Conclusion: There was a significantly elevated expression of Th17 cells and their cytokines ( IL-17 ) in the mice of CIA, which was related to the development of the disease
