摘要
以纯化的狂犬病病毒2μg/mL包被酶标板,将乳糖、麦芽糖、蔗糖和海藻糖分别以5%(W/V)的终浓度加到1%BSA+4%PEG的PBS中,所制备的溶液作为封闭剂,研究其对于37℃贮存板的稳定性,从中选取最佳的封闭剂组合,并研究其对于乙肝表面抗原、猪瘟病毒E2抗原和猪繁殖与呼吸综合征nsp2基因原核表达产物在包被状态下的稳定性。结果显示,以1%BSA+4%PEG+5%蔗糖的PBS溶液的封闭效果最佳,该封闭剂对于乙肝表面抗原、猪瘟病毒E2抗原和猪繁殖与呼吸综合征nsp2基因原核表达产物也具有良好的稳定性。
Purified rabies virus were routinely coated to ELISA plates at 2μg/mL,then only one ingredient of lactose, maltose,sucrose,trehalose at 5%(W/V) was added to PBS which had contained 1% BSA and 4% PEG, the final solutions served as a blocking agents.All were evaluated under 37℃, and then the optimization were screened and further tested on hepatic B surface antigen, swine fever E2 protein and porcine reproductive and respiratory syndrome nsp2 protein.The results showed the solution containing 1% BSA and 4% PEG and 5% sucrose could not only extend rabies virus, but also the others above mentioned antigens under 37℃, so this might be act as a universal coating antigen stabilizer in ELISA procedure.
