摘要
制备聚乙二醇改性壳聚糖(PEG-CS)载体与戊二醛改性壳聚糖载体,并以这两种载体对香菇纤维素酶进行固定化。探讨了载体制备过程中的主要影响因素与最适固定化酶条件,并对两种载体固定化纤维素酶的性能进行对比研究。结果表明,聚乙二醇改性壳聚糖与戊二醛改性壳聚糖固定纤维素酶的Km值分别为1.205、0.626g/L,游离酶的Km值为0.759g/L,两种载体固定纤维素酶的最适温度均比原酶有所提高,且前者的最适温度范围明显变宽;与游离酶相比,固定化酶有良好的操作和贮存稳定性。
Chitosan carrier denatured by glutaraldehyde and PEG was prepared and applied to immobilize cellulase o Lentinula edodes.The immobilization conditions and properties of immobilized enzyme was studied.The results showed tha the Km value of cellulase fixed by PEG-CS and chitosan carrier denatured by glutaraldehyde was 1.205 g/L and 0.626 g/L respectively,and the the Km value of free enzyme was 0.759 g/L.The optimum temperature of the cellulase fixed by PEG-CS and chitosan carrier denatured by glutaraldehyde was improved obviously.However,the range of suitable temperature of the former was wider significantly.Compared to the free enzyme,the operational and storge stabilities of the immobilized enzyme were obviously increased.
