摘要
实验目的:比较不同光能量对大鼠口腔感染性创伤中杂菌的体外灭菌效果。实验方法:选择临床上患有重度慢性牙周炎患者,使用隔湿棉将患者待取材部位进行充分隔湿,使用无菌刮治器取牙齿上近龈缘出的软垢,要求无血。取材后即刻将软垢放置于2ml的0.9%的无菌生理盐水中,于2h内送至微生物实验室,将菌液震荡30s后涂于普通琼脂培养基上,37℃培养24h后用于动物模型的接种。动物模型的建立:Wistar大鼠2只,体重150~180g,雄性,腹腔注射10%水合氯醛麻醉,在上颌第一磨牙腭侧建立1×2mm2创口,将牙龈及其下结缔组织去除,创口止血后,表面涂抹微生物实验室培养的细菌,常规饲养。于2h内送至微生物实验室。实验组的菌液中加入甲苯胺蓝原溶液使其浓度为1mg/ml,避光静置2min,空白对照组不需要加入甲苯胺蓝。实验结果:实验各组均取得了较好的灭菌效果。各组灭菌效果间存在明显差异。光能量48J/cm2照明时灭菌效果最理想,12J/cm2灭菌效果最差。实验结论:光动力疗法能够在体外条件下杀灭大鼠口腔感染性创口出杂菌,并且在光能量为48J/cm2时取得了很理想的灭菌效果,为以后的体内试验和临床研究中光能量的确定提供了参考。
Objective:To compare the sterilization effect of different light intensity on oral wound infections in rats.Methods The materia alba was collected from the periodontal patients with a SRP instrument.Then the materia alba was put into a test tube with 2 ml of 0.9 % sterile saline and sent to laboratory within 2 h.Then the germiculture with the agar medium was performed at 37oC for 24 h.The establishment of animal models:Two healthy male Wistar rats weighing 150-180 g were used in this study.The mice were kept under regular laboratory feeding and boarding conditions.The mice were anesthetized with intraperitoneal injection of 10%hydral to establish the oral wound on maxilla about 1×2 mm2.The bacteria obtained from the laboratory were inoculated into the wound.The TB was used to give final concentations of 1 mg/ml in the experimental but not control group.Results The good bactericidal effect was achieved in all experimental groups.48 J group was the best and the 12 J group was the worst.Conclusion:Our study has demonstrated that a combined TB-PDT therapy can kill bacteria in oral wound in vitro,which provides a foundation for future research.
