从非洲加纳籽提取、纯化5-羟基色氨酸的工艺 被引量:6

摘要 从非洲加纳籽中提取纯化5-羟基色氨酸,在单因素实验的基础上,选择具有显著影响的因素(乙醇浓度、料液比、提取时间)优化球面对称设计优化,得到最佳提取工艺为:80%乙醇以10:1(v/w)的料液比,在70℃下提取2 h,共提取2次,最佳纯化工艺为:提取液以等体积石油醚(60~90°)萃取后浓缩,结晶,结晶物以3%(w/w)活性炭脱色30 min,产品总收率达9.25%,含量超过98%,工艺先进独特,已成功应用于工业生产。 This paper deals with extraction and purification of 5-hydroxytryptophan(5-HTP) from Griffonia simplicifolia in African. Based on single-factor experiment,selected three independent variablels,namely:ethonal volum fraction,solvent-solid and extracting time were affecting factors in extraction.Spherical symmetric design was used for eptimizing the extraction process of 5-HTP.The results showed that the optimum extraction conditions for 5-HTP were as follows:the ethonal volum fraction 80%,ratio of liguid to solid10:1 (v/w),extraction temperature 70℃,extraction time 2 h,total extraction 2 times.The optimum purification conditions for 5- HTP were as follows:extract the extraction liquid with identical volum.Petroleum benzene(60 -90°),decolorize 30 min with the amount of activated carbon 3%(w/w).the total yield of the products about 9.25%,the content above 98%.
