摘要
目的:建立以高效液相色谱法测定风湿Ⅱ号合剂中绿原酸含量的方法。方法:色谱柱为Diamonsil C18(200mm×4.6mm,5μm),流动相为乙腈-0.4%磷酸溶液(10∶90),检测波长为327nm。结果:绿原酸的检测浓度在24.2~193.6μg·mL-1范围内与峰面积积分值呈良好的线性关系(r=0.9999);平均回收率为99.23%,RSD=1.05%(n=9)。结论:本法准确、灵敏、重复性好,可用于风湿Ⅱ号合剂的质量控制。
OBJECTIVE: To establish HPLC method for the content determination of chlorogenic acid in Rheumatoid Ⅱ mixture. METHODS: The determination was performed on Diamonsil C18(200 mm×4.6 mm,5 μm) column with mobile phase consisted of acetonitrile-0.4% phosphoric acid (10 ∶ 90) and detection wavelength of 327 nm. RESULTS: The linear range of chlorogenic acid was 24.2~193.6 μg·mL-1(r=0.999 9) with an average recovery of 99.23% (RSD=1.05%,n=9). CONCLUSION: The method is accurate, sensitive and reproducible for the quality control of Rheumatoid Ⅱ mixture.
