摘要
目的:通过建立血管性痴呆(VD)小鼠模型,探讨补肾强肝活血法治则指导下研制的脑力苏胶囊对血管性痴呆小鼠脑源性神经营养因子(BDNF)表达的影响,为临床治疗VD修复神经功能提供实验依据。方法:双侧颈总动脉反复结扎脑缺血再灌注的方法制备血管性痴呆小鼠模型,造模成功后随机分为假手术组,模型组、脑力苏胶囊高、低剂量组,连续给药14天,治疗结束后观察小鼠一般情况和体重改变,采用ELISA法分别检测VD小鼠脑组织中BDNF含量变化。结果:脑力苏胶囊可升高血管性痴呆小鼠脑源性神经营养因子(BDNF)水平,与模型组比较有统计学意义(P<0.05),其中尤以低剂量组疗效明显。结论:脑力苏胶囊作为体现补肾强肝活血法的有效方剂,可增加VD小鼠脑内BDNF含量,这中作用可能与其改善VD小鼠的认知功能,促进VD小鼠部份神经功能恢复的作用有关。
Objective:Through the establishment of vascular dementia (VD) mouse model to explore the liver and kidney blood circulation while the rule of law developed under the guidance of mental Su capsule on vascular dementia mouse brain-derived neurotrophic factor (BDNF) expression,for clinical Treatment of VD to provide experimental basis for repairing nerve function. Methods:Bilateral carotid artery ligation repeated cerebral ischemia-reperfusion in mice with vascular dementia were prepared model was successful were randomly divided into sham operation group,model group,mental Su capsule high-and low-dose group,continuous administration of 14 days,treatment of mice after the end of the general conditions and body weight changes were detected by using ELISA method VD mouse BDNF content in brain tissues. Results:The brain-Soviet capsule in mice with vascular dementia may be increased brain-derived neurotrophic factor (BDNF) levels,compared with model group was statistically significant (P〈0.05),particularly among low-dose group was significantly effective. Conclusion:The brain-Soviet capsule as the embodiment of the liver and kidney blood circulation law,effective prescriptions,VD increases brain BDNF levels in mice,this effect may be related to the improvement in cognitive function in VD in mice to promote the VD part of the recovery of neurological function in mice related to the role.
