摘要
目的探讨镧离子对神经细胞内游离钙和代谢活力的影响。方法测定Fura2负载的小鼠皮层单个细胞的钙荧光比值,MTT比色法测定小鼠皮层培养细胞的代谢活力。结果0.5~1.0mmol/L氯化镧不影响皮层细胞的静息荧光比值,但明显拮抗谷氨酸和地塞米松及胞外高钾诱发的荧光比值增加;3.0mmol/L氯化镧直接升高皮层细胞的钙荧光比值,并加剧谷氨酸引起皮层细胞代谢活力的抑制。结论皮层细胞内钙受不同浓度镧暴露的影响。
Objective To study the effects of lanthanum on neuronal intracellular calcium([Ca 2+ ]i) and metabolic activity. Methods Cerebral cortical cells enzymatically isolated from neonatal mice were loaded with Fura 2 for measuring the [Ca 2+ ]i fluorescent intensity ratio.The MTT colorimetric assay was carried out to evaluate the cultured cortical cell activtity. Results In isolated cerebral cortical cells,La 3+ 0.5~1.0 mmol/L rapidly reversed the increase of fluorescent ratio induced by L glutamate,dexamathasone or high concentration of potassium chloride,but no effects on their resting fluorescent ratioes were observed.La 3+ 3.0 mmol/L directly increased [Ca 2+ ]i fluorescent ratio and enhanced the inhibition of metabolic activity induced by L glutamate in cerebral cortical cultures. Conclusion The effects of La 3+ on neuronal [Ca 2+ ]i were related with its exposed concentration,La 3+ may directly interfere with the calcium homeostasis in cortical cells and enhance the excitatory neurotoxicity at higher concentration.
