摘要 通过设计多个针对Titin蛋白胞外区多肽序列,用蛋白质微阵列方法筛选抗Titin抗体识别的优势抗原表位,并用ELISA法进行验证.结果显示,在84例MG血清中,51例抗多肽自身抗体阳性,阳性率60.71%.经蛋白质微阵列筛选和ELISA法验证,发现在设计的9条多肽抗原中,P5、P6和P9是优势识别抗原线性表位;进一步发现自身抗体识别的P5与P9多肽表位存在交叉性,并且均能被多肽抗原阻断.结果表明,用蛋白质微阵列方法可有效筛选针对Titin自身抗体识别的优势抗原线性表位. The paper analyzes the screening and identification of the prodominant epitope of Titin.Each peptide-protein couples is dotted on nitrocellulose membrance for arrangement of microarray.Total samples are detected by microarray.The result is validated by ELISA.The results indicates that 51 in 84 cases with MG are detected with autoantibodies against candidate peptides from extracellular domain of Titin showed by microarray and verified by ELISA,positive rate is 60.71%.The paper has further found that P5,P6 and P9 may be the prodominant linearized epitopes in all candidate peptides and there exists overlap between P5 and P9.The results indicate that microarray assay is an important tool for screening and identification of the prodominant epitope recognized by autoantibodies.
