摘要
目的研究氧化苦参碱对银屑病小鼠模型IL-2、IL-10、TNF-α水平的影响,探讨其保护银屑病小鼠的机制。方法采用注射雌激素刺激小鼠阴道上皮组织增生的方法复制银屑病小鼠模型,同时给予不同剂量的氧化苦参碱干预,10d后,取小鼠血清测定IL-2、IL-10、TNF-α的含量,并观察氧化苦参碱对小鼠阴道上皮细胞有丝分裂的影响。结果 80、40、20mg·kg-1氧化苦参碱均可抑制小鼠阴道上皮的有丝分裂(P<0.01),并降低雌激素周期小鼠血清中IL-2、TNF-α的含量(P<0.01);80、40mg·kg-1氧化苦参碱可升高雌激素周期小鼠血清中IL-10的含量(P<0.01,P<0.05)。结论氧化苦参碱对实验性银屑病具有保护作用,可能与其抑制表皮细胞增殖并降低血清IL-2和TNF-α水平,升高IL-10水平有关。
OBJECTIVE To investigate the effect of oxymatrine on the leves of IL-2,IL-10 and TNF-α on mice's psoriasis like animal model of mice and the mechanism of this effect.METHODS The mice vaginal epithelial in estrus cycle was used to produce psoriasis-like animal model by injection of estrogen,and then the mice were treated with three groups of oxymatrine.The levels of IL-2,IL-10 and TNF-α in serum were detected and the effects of oxymatrine on the epithelial cell mitosis were evaluated.RESULTS Oxymatrine of 80,40 and 20 mg·kg^-1 showed remarkable inhibitory effect on mouse vaginal epithelial cell mitosis(P〈 0.01) and significantly decreased the serum IL-2 and TNF-α(P 〈0.01).Oxymatrine of 80 and 40 mg·kg^-1 increased the serum IL-10 in estrus cycle model mice with vaginal epithelium mitosis(P 〈0.01,P〈 0.05).CONCLUSION Oxymatrine has therapeutic effect on experimental psoriasis,which may be associated with its inhibition of the epidermal cell hyperplasia,decreaseing of serum levels of IL-2,TNF-α and the serum IL-10.
