摘要
对酶法转化芦丁后的酶反应产物糖基化芦丁的高效液相色谱(HPLC)测定方法进行了研究。HPLC法的色谱条件为:色谱柱使用Agilent公司ZORBAX Eclipse XDB-C18(4·5mm×150mm,5μm);流动相为V(乙腈):V(超纯水):V(甲酸)=18:81·9:0·1;流动相流速为0·8mL/min;紫外检测波长为254nm;柱温30℃;进样量5μL。运用该方法,在7min之内可以完成样品糖基化芦丁的测定,并且在HPLC谱图上能将芦丁和葡萄糖基芦丁(转入1个葡萄糖单元)完全分离,从而保证测定的准确性。实验结果表明,利用HPLC方法测定糖基化芦丁不失为一种方便、快速、可靠的测定方法。
The HPLC assay method of rutin and its derivatives—transglycosylated rutin in finished enzymatic reaction solution has been investigated.HPLC assay of transglycosylated rutin in the samples was carried out under the following conditions:column,Agilent's ZORBAX Eclipse XDB-C18(4.5mm×150mm,5μm);mobile phase,acetonitrile-water-formic acid(18:81.9:0.1,v/v/v);flow rate,0.8mL/min at 30℃.Transglycosylated rutin(with one glucose unit)and remaining rutin in finished enzymatic reaction solution can be separated completely in HPLC chromatogram.As a result,the accuracy of the assay of transglycosylated rutin can be achieved.Furthermore,the HPLC assay of one sample can be finished within 7min.The results indicated that the HPLC assay method of transglycosylated rutin using 18% acetonitrile as mobile phase was convenient,quick and reliable.
