摘要
以藤黄八叠球菌(Sarcinalutea)为工作菌,用杯碟法测定氨苄青霉素在猪和肉鸡组织中的残留。结果表明,氨苄青霉素在磷酸盐缓冲液、肌肉和脂肪、肝脏和肾脏中的最低检测限分别为0.00125μg/ml、0.00625μg/g和0.0125μg/g。标准曲线的工作范围为0.00125、0.0025、0.005、0.01、0.02μg/ml。浓度在0.02、、0.005和0.00125μg/ml时,变异系数分别是(2.91±0.2)%、(3.39±0.29)%和(8.87±0.28)%。不同浓度氨苄青霉素的回收率在83%~107%。本法快速、灵敏、简便,各参数均符合兽药残留检测要求,适用于测定氨苄青霉素在猪和鸡组织中的残留。
The cylinder plate microbiological assay method with Sarcina lutea was used for the determination of ampicillin residues in tissues of pigs and chickens. The minimal detectable level was 0.00125 μg/ml ,0.00625μg/g and 0.0125μg/g, respectively, for ampicillin in phosphate buffer solution(PBS), muscle and fat, livers and kidney. The standard curve for the following ranges of concentration of ampicillin in PBS: 0.00125,0.0025,0.005,0.01,0.02 μg/ml were obtained. The coefficient of variation value for 0.00125 ,0.005,0.02 μg/ml of the standard curve was (2.91±0.20),(3.39±0.29) and (8.87±0.28)%. The recovery rate was 83%~107% of concentration of 0.02~0.005μg/ml. The result showed that: the microbiological method was rapid sensitive simple. It is suitable for determination of ampicillin residues in tissues of pigs and chickens, with every parameter according with demands of animal drug residues testing.
