摘要
目的改进和完善大鼠急性心肌缺血再灌注损伤动物模型的制备及评估。方法健康SD大鼠随机分为3组:假手术组(Sham组),仅穿线,不结扎LAD;缺血组(Ischemia组),单纯结扎LAD30 min;缺血再灌注组(IR组),结扎LAD30 min,再灌注120 min。每组20只。并从造模成功率、心电图、外周血中LDH、CK-MB的含量测定、心肌病理组织学检查等方面对改进后的模型进行评估。结果改进后的造模成功率达到90%以上,血液动力学、心电图、外周血中LDH、CK-MB的含量测定、心肌病理组织学检查等方式均证实改良后的动物模型成功。结论改进后的方法能大大降低手术难度,提高模型成功率和动物成活率,准确地复制心肌缺血再灌注损伤的实验模型,简化实验操作步骤。
Objective To improve and evaluate the method of establishing rat models of acute myocardial ischemiareperfusion, which had high rate of success and survive. Methods 60 Sprague-Dawley rats were divided into three groups at random: control group, Sham group and improved myocardial ischemia/reperfusion group. The successful rate and the effectiveness of each group were observed. The hemodynamics, eletrocardiogram, Evans blue and TIE staining, myocardial enzyme spectrum and histology results were used for evaluating the modified model. Results The successful rate of three groups were 90%, 100% and 100%, respectively.The hemodynamics,eletrocardiogram,Evans blue and TIC staining,myocardial enzyme spectrum and histology all testified that the models of modified method were results were successful. Conclusion The method of establish rat models of acute myocardial ischemia reper- fusion is simpleand convenient, with high successful rate and survival rate.
