摘要
目的探讨外源性Nogo受体拮抗剂(NEP1-40)在脊髓损伤修复过程中的作用及机制。方法将60只雌性Wistar大鼠随机分为假手术组、模型组和治疗组各20只。后两组均建立大鼠T10脊髓右侧半横断损伤模型,治疗组通过损伤处置管从术后第1天开始每天予NEP1-4015μg溶于PBS缓冲液25μl,连续7d;模型组予等量的PBS缓冲液;假手术组仅去除椎板,不做脊髓半横断,术后不埋管,不用药。分别于术后7、14、21、28d采用Basso-Beattie-Bresnahan运动(BBB)评分法行大鼠行为学表现评分,免疫组化染色法检测受损节段脊髓中NF200的表达。结果治疗组各个时间点的BBB评分均高于模型组,NF200阳性细胞的数目均高于模型组(P均<0.05)。BBB评分与NF200阳性细胞数量呈明显正相关(r=0.937,P<0.01)。结论脊髓损伤后注射NEP1-40可增加NF200阳性神经元数目,促进轴突再生及运动功能恢复。
Objective To evaluate the role and mechanism of Nogo receptor antagonist( NEP1-40) for the repairment of spinal cord injury. Methods Sixty female rats were randomly divided into sham-operated group,model group and treated group with 20 in each . Then spinal right hemisection cord model at T10 were set up in the latter two groups ,the treated group was given NEP1-40 15 μg + PBS 25 μl for 7ds; the model group was given equal equal quantity of PBS. The sham-operated group was removed the neural scute only,and no treatment was given; 7,14,21,28 ds after the operation,the behav- ioral scores were evaluated with Basso-Beattie-Bresnahan ( BBB) scoring method ,the expression of neurofilament 200 ( NF200) was observed by immunohistochemical staining method. Results BBB score and the number of NF200 ( + ) cells in treated group was significantly higher than those in the model group at each time( both P 0. 05) . BBB score was positively correlated with the number of NF200( + ) cells( r = 0. 937,P 0. 01) . Conclusion NEP1-40 can promote the axonal regeneration and recovery of motor function in rats with spinal cord injury.
