摘要
为探索适宜于实验教学的动物组织总RNA的提取方法,采用氯仿处理实验器皿、耗材和水以降低外源性RNA酶对RNA的降解作用。结果表明,操作过程中合理利用氯仿能够有效降低外源性RNA酶的污染;与DEPC水处理效果相比,提取的总RNA均可用于分子生物学研究和RT-PCR扩增,但使用氯仿处理更为安全。
To explore the method of extracting total RNA from animal tissues adapted to experimental teaching,an improved method which used chloroform to treat the utensils,consumables and water to decrease the degradation of exogenous RNA enzymes was established in this study.The results showed that the isolated RNA was intact and suitable for further molecular applications by RT-PCR.This indicates that the method was appropriate for RNA extraction of animal tissues and adapted to experimental teaching.
