摘要
目的分析Pin2/TRF1结合蛋白X1(Pin2/TRF1-interacting protein X1,PinX1)基因在非小细胞肺癌组织中表达,探讨PinX1基因在肺癌发生发展中的作用及其临床意义。方法对56例肺癌组织及癌旁正常组织应用半定量反转录聚合酶链反应(reverse transcription-polymerase chain reaction,RT-PCR)检测其中的PinX1的mRNA表达水平,应用端粒重复序列扩增(telomeric repeat anplication protocol,TRAP)-酶联免疫吸附测定法(enzyme linked immunosorbent assay,ELISA)检测其中的端粒酶活性。结果 PinX1在不同病理类型的肺癌组织中表达无显著性差异,与临床分期和预后之间呈显著相关性,与端粒酶活性呈负相关。结论 PinX1的下调可能与肺癌的发生、发展过程中端粒酶激活有关。检测PinX1的表达水平对评价肺癌预后有重要的临床价值。
Objective To investigate the role of endogenous telomerase catalytic inhibitor Pin2/TRF1-interacting protein X1 (PinX1) gene in the progression of non-small cell lung cancer and its clinical significance.Methods Lung tissue samples and the normal adjacent tissue samples in 56 cases with non-small cell lung cancer were studied.The expression of PinX1 was detected by semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR) and the telomerase activity was detected by telomeric repeat anplication protocol (TRAP)-enzyme linked immunosorbent assay (ELISA).Results PinX1 expression was not significantly correlated with the histological differentiation,but significantly correlated with the clinical stage and lymphnode metastasis of lung cancer.The level of PinX1 was negatively associated with the telomerase activity.Conclusion The down-regulation of PinX1 may function as a telomerase activation factor and thus play an important role in the clinical phenotype and development of lung cancer.PinX1 gene may be used as a molecular marker to determine the malignant degree,metastasis potency and to predict progression of lung cancer.
