摘要
目的建立林蛙不同组织中胆固醇含量的测定方法。方法采用RP-HPLC法,色谱柱为Kro-masil C18柱(150 mm×4.6 mm,5μm),流动相为甲醇-水(体积比为99∶1),流速为1.0 mL.min-1,检测波长为210 nm,柱温为35℃。结果胆固醇的线性范围为8.35-133.60 mg.L-1(r=0.999 9),平均回收率为96.7%(n=9)。结论 RP-HPLC法可作为测定林蛙不同组织中胆固醇含量的方法 。
Objective To establish an RP-HPLC method for the determination of cholesterol in different parts of wood frogs.Methods A Kromasil C18 column(150 mm×4.6 mm,5 μm)was used,using methanol-water(V∶V=99∶1)as the mobile phase;The flow rate was 1.0 mL·min-1;The detection wavelength was set at 210 nm,and the column temperature was 35 ℃.Results The linear range of cholesterol was 8.35-133.60 mg·L-1(r=0.999 9),and the average recovery(n=9)was 96.7%.Conclusions The method is simple,accurate with good reproducibility.It can be applied in quantitative determination of cholesterol in different parts of wood frogs.
