摘要
为探讨自噬在X射线对U251细胞增殖抑制的作用及机制,用X射线处理人胶质瘤细胞系U251细胞,采用WST-1法测定X射线对U251细胞增殖的抑制作用,MDC荧光染色和LC3免疫组化观察辐照后自噬的发生;WST-1法检测3-MA对辐照诱导的U251细胞毒性的影响。WST-1法显示,X射线对U251细胞生长有显著的抑制作用,且此作用呈明显的时间、剂量依赖性;MDC荧光染色和LC3免疫组化。结果表明,X射线可诱导U251细胞发生自噬;X射线照射前阻断自噬使细胞毒性增强。X射线照射能明显抑制U251细胞的生长,并诱导其发生自噬,但自噬对U251细胞可能起到一定的保护作用。
In order to investigate the effects of autophagy on proliferation inhibition in human U251 glioma cells irradiated by X-rays,WST-1 colorimetric assay,MDC staining and LC3 Immunohistochemistry were used. It has been found that X-rays can decrease the viability of U251 cells in a dose-and time-dependent manner by WST-1 colorimetric assay and induce autophagy in U251 cells confirmed by MDC staining and LC3 immunoreactivty. The results showed that X-rays could inhibit the proliferation significantly and induced autophagy of U251 cells.
