鉴别HCV C蛋白上HLA-A2限制的CTL识别表位的研究

摘要 本文先用计算机预测、后用51Cr4h释放分析证实的方法,对丙型肝炎病毒(HCV)核心区蛋白(C蛋白)上,HLA-A2限制性CTL识别表位进行了鉴别。结果发现,2名感染HCV的HLA-A2阳性供血员的外周血单个核细胞(PBMc)对ALAHGVRAL(core150~158)肽段标记的自体靶细胞有溶解作用,杀伤率分别为37.5%和15.8%;用抗CD4mAb阻断后杀伤率无明显变化,而用抗CDSmAb阻断后明显减少,提示该条肽段可能为HLA-A2限制性CTL识别表位。 In this Paper, we attempted to idenop epitopes recognized by the HLA-A2 metricted cytotobo T lymphocyte (CTL)on hepatihs C virus (HCV) core protein utilizing the methed of computer prwhction before conforming by 4 h 31Cr-release assay. The results showed that Peripheral bled mononuclear cells(PBMCS) Obtained fro two HLA-A2 posihve donors who infected with HCV could lyse autolognus target cells theied vitb peptide 'AraHGVRAL(core 150 ~ 158)'. The specific lysis of the cells from the two donors were 37. 5% and 15. 8%, mepechvely. Blocking of the CTL response with and-CD4 showed that the specific lysis had no signifcant decrease. But blocking of CTL resPOnse with and-CDS could abolish the lysis. h indicated that the peptide was the Candidate epitope recwind by the HLA-A2 restricted CTL.
