线粒体KATP通道介导七氟醚预处理对大鼠的脑保护作用 被引量:7

Cerebral protective effect of sevoflurane preconditioning mediated by mitochondrial KATP-channel in rats

摘要 目的探讨七氟醚预处理对大鼠局灶性脑缺血-再灌注(IR)损伤的保护作用。方法雄性SD大鼠60只随机均分为七氟醚预处理组(S组)、5-HD+七氟醚预处理组(HS组)、5-羟葵酸(5-HD)组(H组)、IR组和假手术组(C组)。采用大脑中动脉线栓法缺血2 h、再灌注24 h制备大鼠局灶性脑IR模型(MCAO)。S组于缺血前1 h经半密闭的吸入箱吸入七氟醚(呼气末浓度维持2.4%,持续30 min)。HS、H组缺血前或七氟醚预处理前经尾静脉注射5-HD(10 mg/kg)。再灌注24 h后用ZeaLonga评分法进行神经功能缺陷评分,TTC染色法测大鼠脑梗死容积,免疫组化法测定大脑皮质Bcl-2和Bax蛋白表达。结果与IR组比较,S组神经功能缺陷评分明显降低,脑梗死容积显著减少(P<0.05);但HS、H、IR组神经功能缺陷评分和脑梗死容积差异无统计学意义。S组Bcl-2、Bax表达明显高于HS、H、IR组。结论七氟醚预处理对局灶性脑IR损伤有一定程度的保护作用。其保护作用与mitoKATP通道的激活有关,并可能是通过调控凋亡相关基因Bcl-2和Bax的表达来实现。 Objective To investigate the cerebral protective effect of sevoflurane preconditioning on cerebral ischemia-reperfusion(I-R) injury.Methods Sixty male SD rats were randomly allocated into five groups of sevoflurane preconditioned I-R group(S,2.4% sevoflurane for 30 min),5-HD+sevoflurane group(HS),5-HD group(H),untreated controls group(IR),sham operation group(C).Rats were subjected to 2 hours of middle cerebral artery occlusion(MCAO),which was followed by 24 hours of reperfusion.Neurological deficit of the rats was assessed by Zea Longa's scoring system,infarct size was determined by TTC staining and the expressions of Bcl-2 and Bax in the frontal lobe of cerebral cortex were evaluated by immunohistochemistry.Results Compared with group I-R,the infarct size and neurologic deficit were reduced in group S.5-HD diminished the protection in all two sevoflurane-treated groups,but given alone had no effect on cerebral infarct size and neurologic deficit.The expression of Bcl-2 was significantly higher in group S than that in groups HS,H and IR.Conclusion Sevoflurane preconditioning protects rats brain from cerebral I-R injury,which may be related to activating mitochondrial ATP-sensitive potassium channel and regulating the expressions of Bcl-2 and Bax.
