草甘膦高效降解菌的筛选鉴定及降解特性研究 被引量:6

Isolation,identification and characterization of a highly efficient glyphosate degrading strain

摘要 通过富集分离的方法,从受草甘膦污染的土壤中以草甘膦为唯一碳源进行筛选,共分离出7株对草甘膦具有降解能力的菌株.利用高效液相色谱法对菌株降解能力进行测定,发现QB-7菌株在草甘膦浓度为12 g/L的培养基中,摇床培养7 d后的降解率高达99.34%,具有较高的研究及应用价值.通过电镜的形态特征观察和生理生化试验分析,鉴定该菌株为节杆菌属中的简单节杆菌.并分析了培养时间、pH值、温度、盐度、接种量等几个因素对菌株生长量和草甘膦降解率的影响.结果表明,该菌株在培养时间为5 d、pH为7.0、温度为33℃、盐度为0、接种量为30%的条件下生长较好,并具有较高的降解率. In order to degrade glyposate seven glyphosate degradable strains were obtained from glyphosate contaminated soil by using screening and separation cultural medium method which used glyphosate as the unique carbon source. The degradation rate of glyphosate of the strains was detected by using high performance liquid chromatography method. One of the strains showed potential for further research and utilization with a 99.34% degradation rate which incubating for 7 days in the culture medium with glyphosate concentration by 12 g/L. Based on the photos of electron microscope, physiological and biochemical test results, the strain was identified as Arthrobacter simplex. And factors that can influence the strain growth and the degradation rate to glyphosate, such as incubation time, pH value of the culture medium, temperature, salinity and inoculation amount were also studied in the paper. The resuits showed that the optimum culture conditions for the strain were incubation for 5 d, pH 7.0, incubation under 33℃, salinity of 0 and inoculation by 30%. When the proposed strain grows in this condition, it shows a better growth and degradation rate.
