摘要
目的:应用大鼠胰腺移植模型研究白细胞介素-8/中性粒细胞化学趋化因子(IL-8/CINC)与缺血再灌注损伤的关系。方法:进行同系大鼠异位全胰十二指肠移植,采用血清免疫测定及核酸探针杂交Northernblot方法测定移植后血清IL-8/CINC浓度及移植胰腺组织中IL-8/CINCmRNA的表达。结果:胰腺移植后3h血清IL-8/CINC浓度及移植胰腺组织中IL-8/CINCmRNA的表达呈现峰值。移植后24h胰腺组织内可见大量中性粒细胞浸润。
Objective: The reperfusion injury following pancreaticoduodenal transplantation in rats were studied by measuring serum concentrations of cytokine induced neutrophil chemoattractant (CINC). Methods: Male Wistar rats were transplanted with syngeneic pancreaticoduodenal grafts. The serum CINC concentrations were determined by enzyme linked immunosorbant assay. Expression of CINC transcripts in the pancreas graft was evaluated by Northern blot analysis. Results: The serum CINC concentrations increased and peaked 3 hours after reperfusion (28.4±1.85) ng/ml, decreased gradually thereafter. The serum amylase and lipase concentrations increased and peaked 3 hours after reperfusion. Expression of CINC transcripts in the pancreas graft peaked 3 hours after reperfusion. The neutrophils accumulated in the pancreas graft 24 hours after transplantation. Conclusion: These results suggested that IL 8/CINC was related with the following ischemia/reperfusion injury of pancreaticoduodenal transplantation.
