摘要
目的:通过对本实验室以免疫印迹法确证的HIV-1抗体阳性者WB带型进行分析,总结其内在规律,加深对实验现象的认识。方法:对本实验室自2006-2009年确证阳性者的WB带型进行统计分析。结果:WB带型以全带和次全带为主;gp160和gp120带出现率达100%,p24带出现率达99.91%(1154/1155),而p55带(52.90%,611/1155)和p17带(84.68%,978/1155)出现率较低,其余条带出现率均大于95%;抗pol基因编码的p66、p51和p31三种蛋白抗体检出率均非常高(大于95%),且三者一般会被同时检测到。结论:WB带型及各病毒蛋白抗体出现的内在规律与基因编码蛋白的中心法则和蛋白本身特性密切相关,掌握其规律有助于加强实验室质控,提示早期感染、追踪传染源和传播途经以及临床鉴别诊断等。
Objective:To analyze the western blot(WB) band patterns in HIV-1 positive people and get the internal laws further to enhance the understanding of the experimental phenomena.Methods: All the samples with HIV-1 antibody positive from 2006 to 2009 in our laboratory were collected and the WB band patterns were analyzed.Results: The WB band patterns mainly were total or subtotal.The presence rate of gp160 and gp120 bands were all 100%,and p24 band was 99.91% (1154/1155),while p55(52.90%,611/1155) and p17(84.68%,978/1155) with relatively lower probability.Other bands were detected with high probability more than 95%.p66,p51 and p31 coded by pol gene were all detected with high probability(more than 95%),furthermore,they were detected simultaneously in general case.Conclusion: Certain laws related with the genetic central dogma and the protein characteristics impled in WB band patterns and appearance of the antibodies of viral proteins.The internal laws may be helpful to laboratory quality improvement,early infection implication,tracking sources of infection and transmission way and clinical discrimination and diagnosis ect.
