摘要
从云南农田土壤材料中分离得到一株高产纤维素酶真菌YN-2,对其进行形态及生理生化特征初步鉴定及通过ITS基因片断序列分析后,初步确定该菌株为草酸青霉。产酶条件及酶活力特性分析发现该菌株在培养4 d后,羧甲基纤维素酶(CMCase)酶活达61.50 U/mL,滤纸酶(FPAse)酶活达19.37 U/mL;酶促反应的最适反应温度为50℃;pH值为4.8时,CMCase达52.60 U/mL,FPAse活力达18.37 U/mL。研究发现当固体发酵培养基中添加0.12%的吐温20(Tween 20)对菌株YN-2的CMCase活力影响最显著,在0.10%Tween 20的固体发酵培养基中菌株YN-2的FPA活力有所提高,在其他Tween 20添加浓度的固体发酵培养基中菌株YN-2的CMCase活力和FPA活力均受到不同程度的抑制。
Strain YN-2 with high cellulase-producing activity was obtained from soil of Yunnan farmland,and identified to be Penicillium oxalicum by analyzing its morphology and 18s RNA gene sequence.Experiments of measuring production and analyzing characters of cellulase were carried out after solid fermentation with Strain YN-2.The results showed that the highest activities of the carboxymethyl cellulose enzyme(CMCase 52.60 U/mL) and the filter paper enzyme(FPAse 18.37 U/mL) were obtained after the strain was cultivated with flask shake for 4 d.The CMCase and FPAse activities were optimal at 50℃ with pH 4.8.The CMCase activity was enhanced in the presence of 0.12% Tween 20,and the FPAse activity in the presence of 0.10% Tween 20.However,with other concentrations of Tween 20,both CMCase activity and FPAse activity were inhibited.
